| dc.description | Phosphate solubilizing bacteria was screened from 3 samples such as soil, earthworm intestine and earthworm fertilizer on agar plate of Pikovskaya’s medium (PVK). Ninety-four isolates of phosphate solubilizing bacteria were isolated in this study. The isolated bacteria were further evaluated for phosphate solubilization potential in Pikovskaya broth. The cultures were incubated at 45oC for 7 days. Evaluation of soluble phosphate and pH were performed. Three isolates showed high phosphate solubilization efficiency and has pH in range of 5-6. These isolates are S3, S31.2 and EW32 which obtained soluble phosphate at 246.6, 665.5 and 303.6 mg/L with pH at 5.6, 6.0 and 5.3 respectively. The results of 16s rDNA sequencing analysis showed that S3, S31.2 and EW32 were identified as Bacillus subtilis strain IP6, Bacillus subtilis strain B11 and Klebsiella pneumoniae strain 26 respectively. | |
| dc.description.abstract | คัดแยกแบคทีเรียละลายฟอสเฟตจาก 3 แหล่ง ได้แก่ ดิน ลำไส้ไส้เดือน และน้ำหมักไส้เดือน บนอาหารแข็ง Pikovskaya's medium (PVK) ในการศึกษาอิสระครั้งนี้ พบแบคทีเรีย ละลายฟอสเฟต จำนวน 94 ไอโซเลต จากนั้นนำแบคทีเรียที่คัดแยกได้มาทดสอบประสิทธิภาพในการละลายฟอสเฟตในอาหารเหลวสูตร PVK โดยเพาะเลี้ยงที่อุณหภูมิ 45 องศาเซลเซียสเป็นเวลา 7 วัน ตรวจสอบปริมาณฟอสเฟตที่ละลายได้ และค่าความเป็นกรด – ด่าง สามารถคัดเลือกแบคทีเรียที่มีประสิทธิภาพในการละลายฟอสเฟตสูงและมีค่าความเป็นกรดด่างในช่วง 5-6 ได้ 3 ไอโซเลต ได้แก่ ไอโซเลต S3, S31.2 และ EW32 ซึ่งมีค่าฟอสเฟตที่ละลายได้เท่ากับ 246.6, 665.5 และ 303.6 มิลลิกรัมต่อลิตร ตามลำดับ และค่าความเป็นกรด-ด่างเท่ากับ 5.6, 6.0 และ 5.3 ตามลำดับ จากการบ่งชี้ชนิดด้วยหาการลำดับเบส 16s rDNA พบว่า ไอโซเลต S3, S31.2 และ EW32 คือ Bacillus subtilis strain IP6, Bacillus subtilis strain B11 และ Klebsiella pneumoniae strain 26 ตามลำดับ | |
